His next political role occurred after he was elected National Organiser of the NDC, but John Evans Atta Mills appointed him Regional Minister for the Eastern Region upon taking office in 2009.

Then, Samuel Ofosu-Ampofo moved on to assume his final political position before being chosen as the National Chairman of the NDC in 2018: Minister for Local Government and Rural Development.

During his time in office, he failed to assist the party and John Dramani Mahama in removing the Nana Addo Dankwa Akufo-Addo administration from power in the 2020 general elections.

However, that defeat resulted in a historic victory for the nation: the first ever hung parliament, a split of the exact number of lawmakers, and the Speaker of the House being a member of the opposition.

Samuel Ofosu-Ampofo claimed that one of his accomplishments was that the party had grown more appealing under his leadership in a piece published by the Ghanaian Times.

Additionally, he has drawn comparisons to the historical Jesus Christ.

“The delegates must go no farther than Ofosu-Ampofo as I am ready to lead the NDC to the Promised Land because you do not replace the team that helped the party rebound from a humiliating loss in 2016; I compare myself to Christ when the disciples questioned him about whether he is the Messiah.

Jesus remarked to them, “What more do you want to see? The blind see, the lame walk, and the deaf hear. I am the National Chairman of the NDC, and after our embarrassing loss in 2016, the party was in disarray because it was not appealing. However, I and my staff played important roles in helping the party turn things around in time for the 2020 elections.

He has also claimed that the NDC would have won more parliamentary seats in the 2020 elections, including Techiman South, Tarkwa Nsuaem, Zabzugu, Essikado-Ketan, and others, if not for the employment of state-sponsored security people.

“I have what it takes to return the NDC to power again, but I also believe that, together with my team, we managed to snatch seats that the party had not previously won, including Tema East and Ayensuano, among others, which demonstrates our competence. I am hopeful members, supporters, sympathisers, faithful and well-wishers of the party will renew my mandate to serve for another four years,” he pleaded.
